SUR LES LIGNES DE LUMIÈRE As part of the fight against the SARS-CoV-2 virus, the French National Research Agency launched a call for projects in March 2020 to fund research topics related to the study of this virus. The PROXIMA-1 and DISCO beamlines joined forces with the INRAe and SANOFI to propose the AcceS-Ge CoViD-19 project, which has been selected by the ANR. AcceS-Ge CoViD-19 aims to determine the 3D structures of the constituent proteins of SARS-CoV-2 by in cellulo crystallography. Unlike classical crystallography approaches, the objective in this case is to screen the propensity of each of the SARS-CoV-2 proteins, previously cloned so as to be produced in cells in the laboratory, to crystallise in different sub-compartments of these cells. The initial months of research were therefore devoted to building a plasmid library designed to produce all the SARS-CoV-2 proteins in eukaryotic cells. The second phase made use of all the instruments and experimental methods available on the DISCO beamline, often combined with microfluidics, to determine the best experimental conditions for visualising and localising the crystallised proteins in the cells.
